Mutant Detail

Mutant Detail:
Mutant Symbol12S
Allele SymbolMtnin-1
Gene and Mutation LocationMtNIN (an 11-bp deletion starting at position 1850 at MtNIN locus)
Symbiotic Phenotype BriefCcd- Inf- Nod-
Phenotype Detail12S is Nod-. S. meliloti did not stimulate cortical cell divisions in these mutants, even at a time point in which wild-type plants had fully formed nodules. The mutant also exhibited the characteristic nin mutant phenotype of excessive root hair curling in response to S. meliloti. The formation of infection foci nor threads was observed in the mutant 12S. The mutant formed normal mycorrhizas. Mtnin-1 exhibited a normal calcium oscillatory response following NF treatment. MtNIN is not required for NF-induced gene expression. ENOD11 spatial expression is dramatically altered in Mtnin-1. Out of more than 85 independently transformed Mtnin-1 composite plants, none formed spontaneous nodules.
Single gene targeted?yes
Extra InformationThe F1 progeny of reciprocal crosses between Mtnin-1 and Mtnin-2 were Nod- demonstrating allelism. Mtnin-1 expresses the stronger phenotype of the two alleles.
Mutagenesis MethodFNB
Mutant ClassNod-
Allelismnot allelic to dmi1, dmi2, dmi3, nfp, nsp1, nsp2, hcl. Mtnin-1 and Mtnin-2 are allelic.
The earliest development stage known to be affectedCortical cell divisions
Wildtype LineJemalong A17
