Out of 41 MtCPK3-RNAi transgenic roots screened, no significant differences in root phenotypes were seen when compared with controls (transgenic roots carrying vector sequences or uninfected control roots), and root hair growth or cell size were not affected by our construct. These
transgenic roots were then inoculated with the wild-type
Sm1021 strain. While they were able to produce normal
nitrogen-fixing nodules, the number of nodules was
significantly higher than those in GUS-RNAi control
plants, suggesting that this CDPK isoform participates in the negative control of nodule number in M. truncatula.
